Item type:Article, Access status: Open Access , Biocide testing for the application in the oil and gas industry(2015) Turkiewicz, Anna; Brzeszcz, Joanna; Witek, Wiesław; Kapusta, PiotrThe paper discusses tests for effectiveness of three antibacterial substances for application in the national oil and gas industry. The substances were tested with reference to active cultures of microorganisms. These cultures were isolated, inter alia, from drilling fluids, produced waters and extraction sludge, and then multiplied and used in tests discussed in this publication. The tests demonstrated differences in particular tested substances in respect of effectiveness of their impact on aerobic and anaerobic bacteria and fungi. The results of the work will be used in industrial treatments in order to eliminate biogenic contamination, counteract biodegradation of drilling muds, and also for disinfection of reservoirs designed for storing water and other technological fluids.Item type:Article, Access status: Open Access , Chemical and biological analysis of chromium waste(Wydawnictwa AGH, 2009) Hołda, Anna; Kisielowska, Ewa; Niedoba, TomaszThe paper presents chemical and microbiological analysis (were from qualitative and quantitative point of view) of the collected samples of wastes. The procedure of chemical analysis is based on a quantitative extraction of Cr(VI) from the solid samples and its determination in the extract by the method of flow coulometry by means of apparatus EcaFlow 150GLP, applying calibration by method of model curve. The determination of chromium by flow coulometry is based on utilization of inner electrode coulometric titration. Wydział Geologii, Geofizyki i Ochrony ŚrodowiskaDisposal of waste is now one of the most important contemporary environmental issues and the increasing amount of waste tends to seek new methods of disposal, in addition to burning and costly recycling. Particularly troublesome are the environmental waste polymers, including polyethylene material resistant to degradation. The present study sought to lead to degradation of the chemical structure of polyethylene with the participation of microorganisms. The biological material was isolated from the soil, and the substance has been degraded high density polyethylene (HDPE) in the form of foil. Disinfected samples were subjected to the action of microorganisms suspended in a liquid medium Davis Minimal Broth for 44 days. The medium was a modified chemical composition, so that the main carbon source for the microorganisms was polyethylene. During the breeding experiments the development of microorganisms population was controlled by making measurements of the optical density OD650. The microorganisms were identified as Achromobacter xylosoxidans and the film samples FTIR research showed degradation structure of HDPE.Item type:Article, Access status: Open Access , Ocena i wykorzystanie warunków środowiskowych w procesie wzbogacania kopaliny na drodze (bio)hydrometalurgicznej(Wydawnictwa AGH, 2010) Konopka, Elżbieta; Kisielowska, EwaThese are results of research on the possibility of enrichment uranium-bearing shales (black shales and walchia shales), which at a comparable, trace level of metal-bearing were strongly differed in their geochemical environments, which developed as a result of different compositions of microflora. Research was carried out on the dynamics of growth of micro-organisms most frequently represented in each of the geochemical environments and the associated with varying effectiveness of the leaching. It demonstrated that the enrichment of two radically different raw-shale materials using the biotechnical methods is possible, if taking into account every time changeable, individual and particular method, using the natural biogeochemical conditions of the raw material.Item type:Article, Access status: Open Access , Participation of microorganisms in effluent transformation(Wydawnictwa AGH, 2010) Hołda, Anna; Kisielowska, Ewa; Niedoba, TomaszThe paper presents the chemical analysis as well quantitative and qualitative microbiological analysis of sludge stored in the area of municipal wastewater treatment plant. Furthermore, the influence of microorganisms on sludge properties was investigated. By application of ASA method, the following metals contents were determined: magnesium, calcium, lead, cadmium, copper, nickel, mercury and zinc. Furthermore, the biogenous factors were determined, as amonium nitrogen, overall nitrogen and phosphor. The influence of microorganisms on sludge was also investigated. To this purpose the results given from daily and 3-weekly sludge were compared. The paper presents the problems of possible applications of microorganisms in processes leading to removal of heavy metals from various environments, including coals. The processes occurring with participation of microorganisms as biosorption, bioaccumulation, biotransformation, bioprecipitation, biocrystallization and bioleaching, which can be practically applied in removal of heavy metals from coals connected with their recovery were described in the paper. Wydział Geologii, Geofizyki i Ochrony ŚrodowiskaW pracy opisano metody oczyszczania ścieków, szczególną uwagę poświęcając metodom biologicznego usuwania zanieczyszczeń. Scharakteryzowano osad czynny, jako metodę najczęściej stosowaną i przedstawiono rolę głównych grup mikroorganizmów wchodzących w jego skład. Opisano szczegółowo mechanizmy działania bakterii, które prowadzą do usuwania związków azotu i fosforu ze ścieków. Dodatkowo przedstawiono również możliwości wykorzystania drobnoustrojów do usuwania ze ścieków zanieczyszczeń ropopochodnych oraz metali ciężkich. Wydział Geologii, Geofizyki i Ochrony ŚrodowiskaW pracy przedstawiono ocenę stanu sanitarnego powietrza w Krakowie. Oceny tej dokonano poprzez ilościową analizę liczby mikroorganizmów atmosferycznych w oparciu o metodę sedymentacyjną Kocha. Otrzymane wyniki porównano z proponowanymi dopuszczalnymi ilościami drobnoustrojów w 1 m3 powietrza.
