Wydział Fizyki i Informatyki StosowanejSynchrotron infrared spectromicroscopy and X-Ray Fluorescence spectromicroscopy have been applied to imagine untreated, flash frozen human central nervous system tissue. The hypothesis was that the molecular information available from Fourier Transform Infrared and X-Ray Fluorescence spectromicroscopy would reveal new information about neurodegenerative diseases pathology, specially creatine deposits and their environment. In this study the main focus was on Amyotrophic Lateral Sclerosis. The motor cortex, brain stem, spinal cord and substantia nigra from 6 Amyotrophic Lateral Sclerosis cases and 3 controls were mapped using infrared spectromicroscopy and the results were analysed for protein, lipids and creatine levels. Creatine was found to be focally elevated and formed deposits located in the human Amyotrophic Lateral Sclerosis Central Nervous System tissue. Focally elevated creatine may be a marker of the disease process, indicative of disturbed energy metabolism or inflammatory response to the disease progression. The creatine deposits may therefore be a previously undiscovered disease marker.
