Badania polegały na wyizolowaniu chromo-opornych bakterii z próbki gleby A4 (gmina Alwernia) oraz z próbki gleby ogrodowej, a także określeniu ich przynależności gatunkowej. Do izolacji szczepów wykorzystano agar odżywczy i bulion odżywczy oraz odpowiednie ilości chromianu i dichromianu potasu. Zastosowano następujące stężenia chromu: 1; 2,5; 4; 5,5; 7; 8; 9; 10 g/l (badania wstępne) oraz 7; 8; 9; 10 g/l (badania właściwe). Do określenia potencjalnej przynależności gatunkowej izolatów wykorzystano barwienie metodą Grama oraz obserwacje makro- i mikroskopowe. Z próbki gleby A4 wyizolowano trzy szczepy reprezentujące najprawdopodobniej gatunki: B. firmus, B. cereus, B. circulans, B. pumilus lub B. mycoides. Z próbki ziemi ogrodowej wyizolowano sześć szczepów - trzy reprezentujące takie same gatunki jak izolaty pochodzące z próbki A4 oraz trzy należące prawdopodobnie do gatunku O. intermedium, P. mendocina, P. corrugata lub P. ambigua. Wszystkie wyizolowane szczepy wykazały bardzo wysoką oporność na związki chromu sześciowartościowego (9 lub 10 g/l).
The study involved isolation of chromium-resistant bacteria form soil sample A4 (Alwernia commune) and garden soil sample as well as determination of their generic affiliation. Nutrient agar, nutrient broth and different amounts of potassium chromate and potassium dichromate were used to isolate the strains. The following concentrations of chromium were applied: 1; 2,5; 4; 5,5; 7; 8; 9; 10 g/l (preliminary studies) and 7; 8; 9; 10 g/l (proper studies). Gram Staining as well as macro- and micro-observations were used to determine the probable species of the isolates. There were three strains isolated from soil sample A4 which possibly represented B. firmus, B. cereus, B. circulans, B. pumilus or B. mycoides. In turn, six strains were obtained from garden soil sample - three of them probably belonged to the same species as the strains isolated from sample A4 and three of them presumably represented O. intermedium, P. mendocina, P. corrugata or P. ambigua. All strains were highly resistant to hexavalent chromium compounds (9 or 10 g/l).
