Abstract

Celem rozprawy doktorskiej było opracowanie modyfikacji powierzchniowej stopu Ti6A17Nb, uwzględniając specyfikę warunków pracy elementów potencjalnej endoprotezy: główki i panewki. W pierwszym przypadku, zaproponowano rozwiązanie oparte o warstwy diamentopodobne (DLC) domieszkowane gradientowe krzemem i/lub azotem. Jako potencjalne pokrycie trzpieni endoprotez zbadano nanokompozytowe warstwy chitozanowe oparte o nanocząstki złota (AuNPs) i gentamycynę. W badaniach porównano właściwości warstw DLC domieszkowanych przy użyciu stałego oraz zmiennego przepływu prekursorów domieszki podczas syntezy. Analizie poddano m.in. topografię, właściwości mechaniczne i odporność na zużycie warstw DLC. W przypadku warstw nanokompozytowych skupiono się na wpływie warunków syntezy na morfologię nanocząstek, ich biokompatybilności in vitro oraz określono bakteriobójczość nanokompozytów wobec szczepów bakterii Gram dodatnich i Gram ujemnych. Dowiedziono, że warstwy o gradientowym domieszkowaniu odznaczają się zwiększoną twardością, a w wybranych przypadkach również poprawą właściwości tribologicznych. W przypadku warstw nanokompozytowych stwierdzono, że biokompatybilność nanokompozytów jest zależna od zawartości gentamycyny oraz AuNPs w warstwie. Uzyskane nanokompozyty wykazywały również zwiększoną aktywność antybakteryjną, w porównaniu do warstw zawierających jedynie antybiotyk bądź AuNPs.


The aim of this doctoral thesis was the development of surface modification of Ti6A17Nb alloy. The proposed alloy functionalizations are tailored to the specific work conditions for each element of the prosthesis: the cup and acetabular. In the first case, the proposed solution was based on gradient diamond like layers (DLC) doped with silicon and/or nitrogen. On the other hand, nanocomposite chitosan layers loaded with gold nanoparticles (AuNPs) and gentamicin were chosen as potential stem coatings. The research compared the properties of DLC layers doped using constant and variable flow of the dopant precursor during the synthesis. Properties such as topography, mechanical properties or wear resistance were analyzed. The research regarding the nanocomposite layers was focused on the influence of the synthesis parameters on nanoparticles’ morphology, their biocompatibility in vitro and assessing the antimicrobial activity of the nanocomposites against Gram positive and Gram negative bacterial strains. The results showed higher hardness and, in some cases, improved tribological properties of gradient DLC layers. In regards to nanocomposite layers, biocompatibility of the nanocomposites depended on gentamicin and AuNPs loading in the layer. Produced nanocomposites also showed increased antimicrobial activity, compared to the layers loaded with only antibiotic or AuNPs.
